Graduate student at Northwestern University studying bio sciences and BIGG DATAAAAA; still know my electrical engineering though.

  • Learning:
    • Python (+ wxPython, PIL Image, others),
    • PowerShell
    • Java (on Android platform)
  • Know C on embedded well (what's a file? stream? :P)
  • Used to know HTML (sort-of now), CSS (less-so...), JScript (no.) At least I was able to forget most of this stuff just in time for AJAX to become useful. (Because it's totally different from DHTML...)

I like stout, esp. oatmeal stout. Gin and tonic if we're talking mixed drinks. Also like Portal (hell, all Valve games), and would kill a bus full of baby nuns with fire if Gabe would release Half-Life 3 already.

Ampicillin degrades by hydrolysis (albeit very slowly if not catalyzed), difficult when in dry, much easier when in solution.

@nico I don't know, clearly someone wants them or they wouldn't be offered. I've only done a couple PCR reactions, and only with Phusion (Pyrococcus derived; "50X" fidelity)

It certainly has been, that's just an example demonstrating it's ability to be artificially cultured. Biotech companies that produce native Taq probably have all sorts of tricks and optimizations that they'd never share.

The question of them being alive is moot; prions are ultimately dependent on DNA for their propagation as the host must generate additional substrate for them to turn over.

There are protocols on how to culture T. aquaticus, but I don't see any on how to get any decent expression of Taq pol. Maybe it expresses extremely well anyways? It still seems asinine that companies charge a mere ~10% price premium on the native vs. recombinant protein; probably why a startup that is sharing our building is able to carve themselves out a market.

Would be nice to know why some of my T7-promoter plasmids produce ~100 mg protein/L of culture and others maybe 2-3.
