Could you attach a picture of the gel? Also, how many times was this experiment repeated? I have used different volumes many times with no difference in efficiency (but with Phusion polymerase)

I also had a feeling that the protein concentration should be more uniform, especially for the same protein (GFP in this case). I also imagined it would not be lower than 40%.....
