comment How can I make sure my UV-purificator kills bacteria?
OP is looking to "check if purificator kills most of the bacteria" - viable cell count is probably overkill for that. I suggest a simple comparative approach. Dead bacteria don't behave like live bacteria, so one only need compare the chemical content of the water containing "possibly dead" and "known live." Say there's no difference - it wouldn't make sense to do a cell count at all. You would expect the known live bacteria to do more than a dead one, thereby changing the chemistry of the water.

comment How does the brain manifest and realize the intention to resist sleep?
OP is asking about how the brain resists sleep, not what the results would be.

No. See this answer to 'How fast do different organs turn over cells?' and this answer to 'Why is an HIV infection considered “incurable”?'.

comment Producing a genetically modified animal with cell walls
The short answer is it's very complicated. Plants devote a significant portion of their genomes to cell wall construction and integrating that into an animal, I imagine, would be like integrating a helicopter into a battle tank.

comment Enrichment of slow growing microbes: Large Test Tubes as an alternative?
Yes, that was my thought. Obviously not ideal since you'll have to be careful, but if that's all you have on hand it's what I would do.
