Answering to the significance of contribution to A660 by casein, we performed a spectrum check and then proceeded with A660. |

Growth kinetics of E.coli on media containing lactose and casein The concentration used was 2% w/v. And no, I don't think that it contributed significantly, for the growth curve we got was sigmoidal. The inoculum was a pure culture of E.Coli obtained from degrading raw milk. I too had similar thoughts about the negative value being impertinent to the growth of cells. though I was thinking on the lines of cell acclimatization and lag phase proteolytic activity. And obviously cells will use glucose and lactose present in the medium first which could be inferred from the media without casein giving a growth curve which closely following the standard sigmoidal. |
