I am a science enthusiast. I plan to work on biomolecules, interested in Structural and Computational Biology. And I am here to take in every bit of information I get!!! Greedy eh...!!! Yeah... I am...!!! ;)

comment Growth kinetics of E.coli on media containing lactose and casein
The culture was Ampicillin and penicillin resistant and tetracycline susceptible, if it helps.

I just want another pov that whether it was the casein which could have played the role in getting the lag phase negative values. Answering to the significance of contribution to A660 by casein, we performed a spectrum check and then proceeded with A660.

The concentration used was 2% w/v. And no, I don't think that it contributed significantly, for the growth curve we got was sigmoidal. The inoculum was a pure culture of E.Coli obtained from degrading raw milk. I too had similar thoughts about the negative value being impertinent to the growth of cells. though I was thinking on the lines of cell acclimatization and lag phase proteolytic activity. And obviously cells will use glucose and lactose present in the medium first which could be inferred from the media without casein giving a growth curve which closely following the standard sigmoidal.

I'm only concerned about the aspect where it shows the negative values. And I think that it must utilize casein (to a certain extent) because I isolated it from raw milk.

comment Alternatives to TBE buffer for denaturing polyacrylamide gel electrophoresis of DNA and RNA?
sure.. try it once... and run any marker, along with running the same marker with TBE if possible, if you can to check for possibilities...
