@nico, I think I have pointed out that viruses do not qualify under the above definition. Likewise as mentioned in biology.stackexchange.com/a/96/389, prions are dependent on host factors. Citations will come after I finish my experiments.

Well at the moment, there isn't a question related to what is the definition of life. Meanwhile, I think that most evolution of life specialist will say that no, cells are not the basic unit.

Not related to your question but what voltage are you running your gels? It seems as if your having all sorts of issues with over-heating.

I think the folklore is influenced by how much nucleic acid you are working with. As @Daniel Standage suggests, some conditions wouldn't be influenced by anything.

How strange. I'm looking at the crystal structure (1QLN) and there aren't a lot of cysteines near the active site. Will have to take a look at those papers.

I don't think that this is necessarily correct. The beta-lactams doesn't inhibit the synthesis but rather the reorganization of the peptidoglycans. The rigidity of the now static wall then prevents cell division.
