Alternatives to TBE buffer for denaturing polyacrylamide gel electrophoresis of DNA and RNA? I've used Lithium Borate with an agarose gel and high voltages (>200). Didn't turn out as well as advertised the few times I tried it though - worse than TAE. If I had to try it again I would definitely lower the voltage. |

Is there a cheap and easy way to check for potentially unsafe UV exposure? That does seem obvious after looking at products available in a google search. Maybe one the reusable ~6 level color changing sensors would be adequate or even one of the ~$30 handheld digital models. Spending several hundred dollars is out of the question though. I was just hoping someone would say, oh yea all labs in the world use this cheap x sensor to warn us of excessive exposure (analogous to radiation badges). |

X chromosome “weight”? Girls take note, this is not a valid excuse. Seriously though, this difference is exploited in some (albeit minimally effective) techniques to sort sperm for gender selection. |

Does an annealing temp higher than primer's Tm contribute to primer dimer? Ironically this is one of the few times were I didn't design the primers and used what was mentioned in numerous papers. |

Does an annealing temp higher than primer's Tm contribute to primer dimer? I like your example and calculation. I guess I didn't think Tm was so polymerase specific before. Coincidentally it ends up at the same annealing temperature that I tried though. Unfortunately the taq I am using gives very little information (and not an online calculator) in regards to choosing temperatures. I am close to ordering a new mastermix anyway, so I will likely try a different brand then. But as for my current problem there are enough variables that I plan to try the same annealing temperature again and if that fails completely then I may look at what @jp89 suggested also. |

Alternatives to ethidium bromide for staining small nucleic acids? Interesting. I haven't done a comparison so I can't speak one way or the other. But I do remember a similar article sometime back saying a similar thing (EtBr is not as bad as you think, and SYBR Green really isn't a "safer alternative" as advertised). |
