Jul 1, 2013 at 8:09 comment added Gianpaolo R Using only two self-annealing oligos as both primers and template, you will just fill the in withouth amplification. The reaction will occurr even withouth thermal cycling, which renders the presence of thermocycler unuseful. Perhaps the worst experiment ever to do to test a thermocycler :-) Thanks @Alan Boyd for pointing it out. Btw, oligos are normally shipped lyophilized at room temperature in small tubes inside normal envelopes, ordering 2 or 4 will not affect shipping costs.
Jul 1, 2013 at 3:23 comment added WYSIWYG yeah it can be done using two oligos.. but it is better to not have low complexity sequences like AAAAA and TTTTT etc.
Jul 1, 2013 at 1:31 comment added DucFabulous Hello, why you removed the edited answer ? It's look like a good idea, can it's possible if i using only 2 oligo 3'-NNNNNNNNNNNNNNNNAAAAAA-5' and 5'-TTTTTTTTNNNNNNNNNNNNNNNN-3' ?
Jun 30, 2013 at 17:39 comment added Gianpaolo R @Alan, thanks I removed from the answer. Fill in, will not be useful to test the thermocycler.

Jun 30, 2013 at 15:04 comment added Gianpaolo R I edited my answer: you can do PCR with two oligo without any other DNA template. This is the simplest way I think.

improved the answer with more details
Jun 30, 2013 at 14:30 comment added DucFabulous I think my PCR machine is cheaper, faster, special because i using a motor to bring the tube from 95 o C to 65 o C just in a few second. If you like my PCR, i wil sell it for you cheaper than the OpenPCR ;)
Jun 30, 2013 at 14:27 comment added DucFabulous Well, amazing idea ! But 4 oligo could make more cost than 2 oligo because i need to ship oligo from foreign country so price is an important factor in my experiment. Anyway thank you for that idea. I'm finding another way simpler than that when DNA can be found in any living organism.
